Palmitoyl Tetrapeptide-7

Cosmetic / Anti-Inflammatory Peptideapproved

Also known as: Pal-GQPR, Rigin, Palmitoyl Tetrapeptide-3, Matrixyl 3000 component

A lipopeptide used in cosmetic skincare that reduces inflammation-driven skin aging by suppressing IL-6 and other inflammatory cytokines, often paired with Palmitoyl Tripeptide-1 in anti-wrinkle formulations.

Overview

Palmitoyl Tetrapeptide-7 is a synthetic lipopeptide consisting of four amino acids (Gly-Gln-Pro-Arg) conjugated to a palmitic acid chain for enhanced skin penetration. Originally known as Rigin, it was developed as an anti-inflammatory cosmetic peptide that targets immunosenescence-driven skin aging. As skin ages, chronic low-grade inflammation ('inflammaging') accelerates collagen breakdown and impairs repair. Palmitoyl Tetrapeptide-7 suppresses the secretion of interleukin-6 (IL-6) and other pro-inflammatory mediators, reducing this destructive inflammatory cascade. It is most commonly found in combination with Palmitoyl Tripeptide-1 in the trademarked complex Matrixyl 3000, one of the most popular anti-aging peptide combinations in clinical skincare. Together, they both reduce collagen destruction (via anti-inflammation) and stimulate new collagen production.

Mechanism of Action

Palmitoyl Tetrapeptide-7 works by suppressing the production of excess interleukin-6 (IL-6) by keratinocytes and other skin cells. IL-6 is a key driver of chronic inflammation in aging skin that upregulates matrix metalloproteinases (MMPs), enzymes that degrade collagen and elastin. By reducing IL-6 levels, the peptide slows the inflammatory destruction of the extracellular matrix. The palmitoyl (C16 fatty acid) lipid tail enhances penetration through the stratum corneum and improves cellular uptake. The tetrapeptide sequence mimics fragments of immunoglobulin G (IgG), which may explain its immunomodulatory properties.

Pros & Cons

Targets the root cause of skin aging (chronic inflammation) rather than just symptoms

Excellent safety profile — used in cosmetics for over a decade with minimal adverse reports

Synergistic with Palmitoyl Tripeptide-1 — combined as Matrixyl 3000 for dual-action anti-aging

Non-invasive topical application with no injections required

Effects are subtle and cosmetic-grade — not comparable to injectable peptides or medical procedures

Limited peer-reviewed clinical data — most evidence is from manufacturer-sponsored studies

Concentration in commercial products is often very low and may not match study doses
