Overview

Palmitoyl Tripeptide-1 is a synthetic lipopeptide consisting of the tripeptide sequence Gly-His-Lys (GHK) linked to a palmitoyl (palmitic acid) fatty acid chain. The GHK sequence is a naturally occurring fragment released during collagen breakdown that signals the body to produce new collagen — it essentially mimics a 'wound signal' that triggers repair processes. The palmitoyl group enhances skin penetration and stability. This peptide stimulates production of collagen types I, III, and IV, as well as fibronectin and glycosaminoglycans (including hyaluronic acid) in the dermis. It is often combined with Palmitoyl Tetrapeptide-7 in the commercial product Matrixyl 3000 for synergistic anti-aging effects. Palmitoyl Tripeptide-1 is one of the most clinically supported cosmetic peptides available.

Mechanism of Action

Palmitoyl Tripeptide-1 activates collagen repair pathways: (1) The GHK fragment mimics collagen breakdown products, signaling fibroblasts to increase collagen synthesis; (2) Activates TGF-beta signaling cascade in dermal fibroblasts; (3) Upregulates genes for collagen I, III, and IV production; (4) Stimulates glycosaminoglycan (GAG) synthesis including hyaluronic acid; (5) Promotes fibronectin production for extracellular matrix integrity; (6) The palmitoyl chain enables penetration through the stratum corneum lipid barrier; (7) Mild wound-healing signaling without actual tissue damage.
